Anti -Hcv Lectin from Egyptian Pisum sativum
نویسنده
چکیده
Lectins are carbohydrate binding proteins expressed in plants, animals and microorganisms and have been used to probe the surface properties of a wide range of prokaryotic and eukaryotic cells. The implication of some lectin molecules in several physiological processes has been claimed. The aim of this work is to purify the lectin from Egyptian pea (Pisum sativum) seeds and study its biochemical characterizations and potential as pharmaceutical compound. Egyptian Pea lectin seeds was purified by ion exchange chromatography (DEAE column) followed by Sephadex G 100 Column. Purified lectin showed Hemagglutinating activity with human and rabbit erythrocytes and appeared as one band of 32 kDa on SDS-PAGE gel, also amino acid analysis showed high content of glutamic and aspartic acids but very low levels of sulfur amino acids. The purified Egyptian pea (Pisum sativum) lectin showed high effect on HCV replication at concentrations of 20, 40 and 80 μg. Therefore, pea's lectins might consider as one of the natural compounds that can be used in designing a safe and effective anti HCV therapy.

Isolation, characterization and production of a new recombinant lectin protein from leguminous plants
Lectins are present in microorganisms, plants and animals and have attracted great interest due to their varied physiological roles in cell agglutination, anti-tumour, immunomodulatory, antifungal and antiviral effects. Legume lectins are important to the pharmaceuticals but they are produced in low amounts in the plant seeds. Moreover, the genes controlling these proteins are conditionally act...
متن کاملDesign, expression, and crystallization of recombinant lectin from the garden pea (Pisum sativum).
The propeptide form of the lectin from the garden pea (Pisum sativum agglutinin) has been expressed in Escherichia coli by attaching its cDNA to an inducible promoter. By a number of criteria, including the ability to form dimers, hemagglutination titer, Western blot, and enzyme-linked immunosorbent assay, the resulting propeptide molecule is virtually indistinguishable from the mature proteoly...
متن کاملIn vitro callus induction for determination of lectin activity in pea (Pisum sativum L.) variety (AP-1)
From 8-10 days old seedlings of Pisum sativum L. (variety AP-1), various parts root, cotyledon as well as leaf were separated to use as explants. Root and cotyledonary explants did not show any response on MS (Murashige & Skoog) media supplemented with 2, 4-D at a concentration of 5 ppm. However, leaf explants showed callus formation on this media. The callus so formed was subcultured on MS med...

Asparaginase was extracted from plant parts of Pisum sativum subspp. Jof collected from a field crop. Asparaginase activity was detected in seeds, stems and leaves extracts. Enzyme activity was higher in seeds extracts (30.0 U/ml) compared with leaves extracts (26.4 U/ml) and stems extracts (16.1 U/ml), respectively. Optimum conditions for the activity of crude asparaginase extracted from plant...
متن کاملSerum Mannan-Binding Lectin in Egyptian Patients With Chronic Hepatitis C: Its Relation to Disease Progression and Response to Treatment
BACKGROUND Chronic hepatitis C virus (HCV) infection is a major worldwide public health problem. Egypt has the highest prevalence of adult HCV infection in the world, averaging 15%-25% in rural communities. Mannan-binding lectin (MBL) is a liver-derived pluripotent serum lectin that plays a role in the innate immune system of the host. It is an acute-phase protein that is involved in the activa...
